How can I connect Shimmer3 device via bluetooth to myRio directly without using the PC.

I'm using PmodBT2 module

It would probably be best to consult the documentation for the PmodBT2 module to determine how you should connect it to the myRIO, and how it communicates. We can't really offer specifics on how to connect a third party device to our device through another third party device, since we are not familiar with those other devices.
