How to select the which handshake lines

I have a PCI DIO 32 HS with 2 groups of handshake lines.
I'd like to use the Req1/Ack1 as handshake, the other
two as normal IO lines. Upon looking at the examples,
it somehow escaped me where that would be set with
what command. Now the level ack mode appears to be
sensitive on both pairs.

Hello;

The control lines of the 32HS can be used as data lines as long as none of them are used as control lines.

Since you are using 2 of the lines as control lines, you can't use the others as data lines.

The DAQ 653X User Manual p2-3 states :

Using Control Lines as Extra Unstrobed Data Lines
[ 2 Controllers, bla, bla ] plus :
If you're not using Group 1 and/or Group 2 as timing
controllers, ... ,you can use their control lines as
extra data lines.

I tend to interprete this not as strong as your
answer. Meaning the other pair should be useable.

Could you please verify ?

Yes, the manual is correct. If you're only using one group, not both, for handshaking or pattern I/O, then the four control lines for the other group are available for unstrobed I/O. The software implementation is described on pp. 2-5 and 2-6 of the current user manual at http://www.ni.com/support/.
