We are using the dig buf handshake in(8255) VI to read 6 lines from a PCI 6503 card. We get one of two errors: either "this device does not support this action" or "the specified channel is in use". What am I doing wrong?

We've set the group to 0, but not fully understanding the definition of "group" this seemed the only logical choice.

Hello;

You didn't include on your posting what OS you are using there.
One thing to consider is that DAQ Event, and DAQ Occurrences are only supported under Windows platforms; so, if you are using other OS than Windows, that might be the cause of the error.
