How to determine this mean frequency ???

I need to measure a striation frequency (from a pict of a cell) and I don't manage to do that...

I really looked like you were on the right track. We did something similar using FFT on the line extracted data. Are you manually selecting the line? What does the raw data of the extracted line and FFT show?
