How to use a SubVI as a trigger for main VI?

You would typically not use the run button in an application.  Your application should already be running, perhaps in idle mode waiting for a trigger from the 6321.

Ok that's just the way the professor I'm doing research under recommended. Where would the trigger from the 6321 come from? I have attached two programs, the "y-galvo" is a program that another student created, which is supposed to be SubVI #1 in that diagram I posted. The other program I attached is meant to be triggered off of the first SubVI and create 500 digital pulses.

Well, you can just use a push-button on the front panel to trigger your measurement.  Or, if your wanting something automated, you would have to determine what your trigger event is.
