However, the 2016 FPGA Module readme says it is not compatible with windows 10!

As you can see in your first link, the FPGA module works with Windows 10. So you can create FPGA VIs on Windows 10.
The Xilinx Tools, on the other hand, do not officially support Windows 10. So you can't compile your FPGA VI on a Windows 10 machine (again, officially. There might be ways to make it work as has been discussed previously).
You could install the Xilinx Tools on another machine and compile there, or use the NI cloud compile service.
