Verify dot mark presense and absence at defined location

I am having a difficult time finding a way in vision to verify the presense and absense of dot mark in the image at defined location.  I am attaching image.

  Can anyone help?

Your dot marks can hardly be seen (it took me a while myself to find it). Can you enhance the image (contrast enhancement) before thresholding the image using red plane exraction.

I tried enhancing the image but no result from it. In thresholding the whole surface looks similar and could not able to extract the dot .

 help me to verify the presence and absence of dot.
