My guess is that when you talk about EMG you refer to electromyography; so after researching a little bit about this sensor I can see the output is an electrical signal related to muscle activity. The card that you have has 64 single ended analog inputs or 32 differentials AI, in your case the connection you want to choose is “differential”. If we start from the sensor to the 6071E you will have; the EMG sensor that "should" have two wires, each of those wires will go connect to a terminal block. The places you are going to connect these cable to the PCI-6071E page 2-24 will show you how the differential are made. One of the best resources we have is “Measuring Floating (No referenced) Sources”. Something to consider is since the output of an EMG is a very low voltage you will have to make sure that your voltage range is change to ensure accuracy.
