a current source in parallel to AO voltage and both of them are connected to a resistor. Will it damage the AO pin?

It depends....
- On the AO voltage.
- On the current from the source.
- On the resistance.
It is probably not a good idea. What are you trying to do?

Actually I have a IC chip who has 2 pins that are connected to thermostat resistor. They are used to measure the flow rate. The voltage difference between 2 pins internally is measured by IC chip.
I try to simulate/model the difference between those 2 pins. I keep one of pin voltage constant and adjust slightly on the other. At the beginning, both pins have same current flowing through its resistor. I connect the voltage source and apply a new voltage to one of pin. It generates the voltage difference. Is it a right way to do? or Any other good suggestion.
thanks

I am having some difficulty understanding how you have everything connected and exactly what you are trying to do. Can you post an image (.png preferred) of the wiring or schematic diagram? What model is the IC?
