I am trying to generate a digital pattern ( to turn on/off ) sequentially LED's while acquiring analog data on one channel. Using SE 2.0 and a 6251 USB. Can I generate the pattern while acquiring analog data or is it better to acquired data, change the pattern and acquire more data. The latter seems to lead to a large number of "add steps".

Although the help says I should be able to select any type of digital trigger for a digital output, the choice is always grayed out.

Any advise or examples would be welcomed.

You can put both the Digital Generate Step and the Acquire Step in a Sweep.  Configure the digital step to be line output and add the desired lines.  Then you can sweep the value of each line.  The same sweep step can sweep all of the lines together.  I would probably configure the swep to be in list of points mode so that you can specify the value of each line at each iteration.  Let me know if you need an example of this.

Can I assume this is true for rev 2.0 and 2.5? Is sweep a term I'll find in the help menu?

The sweep is a step you can drop.  It is under the execution control palette.  To sweep the Digital Line you need to have DAQmx 8.6 which shipped with SignalExpress 2.5 but can be downloaded separately if you have 2.0.
