You can make the two buffers different sizes. You can think of the Buffers as two different buckets.
DMA Host to Target: In the background LabVIEW will try to empty the Host (PC) and keep the Target (FPGA) filled.
DMA Target to Host: In the background LabVIEW will try to empty the Target (FPGA) and keep the Host (PC) filled.
