Thank you for letting us know what device you are using. this device is listed under support devices for NI-Serial 3.6; and the backwards compatibility for the functions you are using shouldn't be an issue either.
I don't believe I/O Trace captures serial data; that is what Portmon would do.
Are you only upgrading the drivers or also the OS?
I ask because there are some specific instructions that need to be followed when upgrading from an older driver to a newer one.
Please take a look at the readme Release Notes.
After upgrading to NI Serial 1.6 did you disconnect and reconnect the card? This updates the firmware.

The driver 1.6 works correctly. I upgraded another machine to 3.6 and it does not function. If I change the comport to COM1 (PC comport), the code works correctly. If I use COM 3 (port 1 on 8420 card) it does not. I have the correct base address for the ports. We would like to upgrade drivers but need to resolve this issue before we upgrade the 40+PXI racks on our production floor.

Could you please provide some step-by step details of what you did while upgrading? Have you diconnected and reconnected the card in order to update the firmware? Also, are you able to run a shipping example from the NI-Serial 3.6 driver install successfully?

Just to verify, to upgrade the card you need to remove it while the rack is powered, then plug it back in?

Here is the proper upgrade procedure as listed in the serial installation guide.
1. Log on as Administrator or as a user with administrator privileges.
2. Insert the NI-Serial Software for Windows CD (or run the setup wizard from the download)
3. Click Install Software and follow the onscreen instructions.
4. Power down the computer, install your serial PCI, PCI Express, PXI,
or PXI Express hardware, and power on the computer.
5. After Windows detects your hardware, select Start»Programs»
National Instruments»NI-Serial»Troubleshooting Wizard.
6. The Troubleshooting Wizard window appears. This application
verifies the software and hardware installation and sequentially
tests each NI serial port.
7. Connect the cables.
