Hello,
Depending on what 5B backplane that you have, you can connect two E series cards to the 5B backplane via two NI 2056. I am assuming that you are connecting the two NI 6071 E series cards to two SC 2056, where each NI 6071 has its own SC 2056. The two SC 2056 are then going to connect into the two 26 pin connectors (P1 and P2) located on some of the 5B series backplanes (some of the backplanes only have one 25 pin connector). This setup is possible. In fact, notes related to these P1 and P2 connectors note that P1 and P2 are wired in parallel for daisy chaining up to 4 boards.
