I don't know that there really is a 'Best Practices' for this. There are examples: http://zone.ni.com/reference/en-XX/help/370052H-01/tssuppref/infotopics/property_loader/ that you can go off of.
Also, in the TestStand Reference Manual in Appendix C there is a section about it. This should help as well.

I've found the easiest way for me to start using the property loader is to let TestStand do the work. By that I mean using Tool>>Import/Export properties and create your .xls spreadsheet there. In this manner you can create a properly formatted spreadsheet that the property loader step can understand. When configuring your property loader step just point it at the .xls spreadsheet you created using the export properties tool and make sure you use the same start and end markers.
