Have you tried hooking up any other VIs to the initialize VI? This can determine if the problem is caused by the initialize block or the Prepare Lambda Scan.VI
I would also use the highlight execution (the light bulb on the block diagram) to track the progress through the block diagram to make sure the VISA resource being passed is what you expect. I see a couple things in the initialize that could be causing a problem, but since I do not have the instrument, I can not tell. Try running and tell me what you get on Instrument handle out from the getting started VI.
