Regarding the PXIe-4139 SMU, it is a single-channel module. Do any of u had an idea that:
Is it possible to modify or configure the PXIe-4139 to support two channels and would there be any impact on the module’s performance or specifications?

What do you even mean? it is 1-channel by design, you can't just add a new channel out of thin-air. This is analogous to asking can you make a 1-SIM mobile phone to support 2-SIMs.
If you mean, you want to share the same channel for multiple loads, sure, as long as they need the same sourcing condition and its under the current limit of the instrument.
