Thank you for your post. Which manual did you read? There is a special OEM guide, you can find this guide also under the following link;

One page 9 you will see that the 6251 OEM contains two headers; one for digital I/O and one for the analogue input / output. Please not that you must provide the DAQ unit with power through a external power supply. Please refer to page 17 for more information about the 34 pin and 50 pine header.
The following manual tels you more about how to connect your signals to the DAQ device. For example how to connect a single ended signal to your analog input channel (chapter 9).
